Staining procedure for thin blood film 1. When the blood smear is totally dry, arrange the slides on the slide rack, with the blood smear facing front. The slides must not be in contact with each other or else the stain will be flow off. 2. Cover each slide completely with 1-2ml of Leishman’s stain (undiluted) using Pasteur pipette and leave for around 45seconds. 3. Add the buffer. Mix the stain by blowing gently to and fro. 4. Leave the slides to stain for 17 minutes. (The time can be modified if necessary, with different batches of stains – from 12 to 20 minutes) 5. At the end of 17 minutes, wash the stain off by pouring tap water onto the slides from one side and rub off the remaining stain at the back of the slides by using a sponge. 6. Put the slides at the slide stand for air dry. Staining procedure for thick blood film 1. Place the thick smear slide flat on a staining rack. Carefully overlay the entire smear with demineralized water. 2. Allow the water to evaporate and the slide to air dry completely. DO not heat or disturb the slide during this phase. 3. Dip the dried smear in Giemsa Plus fixative 5 times, 1 second per dip and drain the excess. 4. Then, dip the slide in Giemsa Plus Reagent A 5 times, 1 second per dip. Drain the excess. 5. Lastly, dip the slide in Giemsa Plus Reagent B 5 times, 1 second per dip. 6. Rinse the smear with distilled water and allow to air dry.
